Anthrax lethal factor inhibition

Time lapse movie showing J774A.1 macrophages treated anthrax lethal toxin along with the metalloproteinase inhibitor GM6001 (left panel) or vehicle control (right panel).  The movie covers a time period from 2 hours to 6 hours after the addition of toxin.  Cells without drug show extensive necrosis while those receiving the inhibitor appear completely healthy.

LF-peptide complex (PDB 1PWW)

LF-GM6001 complex (PDB 1PWU)

LF-thioacetyl inhibitor complex (PDB 1PWQ)

Anthrax lethal factor (LF) in complex with substrates and inhibitors were solved in Robert Liddington’s lab at the Burnham Institute by Thiang Yian Wong with help from Robert Schwarzenbacher.

Pim-1-inhibitor-peptide complex (PDB 2BIL)

The Pim-1 kinase in complex with the optimized Pimtide substrate was solved by Alex Bullock in Stefan Knapp’s group at the Structural Genomics Consortium, Oxford.
